Phase 1b/11 study of APR-246 in combination with chemotherapy in platinum sensitive relapsed high grade serous ovarian cancer

This novel agent has 2 anticancer actions - firstly it reverses the unfolding of the tumour suppressor p53 and secondly it reduces glutathione levels and inhibits thioredoxin reductase. Both mechanisms may be relevant to targeting tumours with p53 alterations, detected in over 50% of human cancers, by promoting apoptosis. Secondly enhancing sensitivity to cytotoxic agents including cisplatin and radiation by altering the REDOX status of the cell.
