Myotubularin (MTM) family is a large group of dual-phosphatases
(DSP) formed for sixteen proteins. MTM family is dividided in two subgroups
of active and inactive enzymes. All of them present a PHG domain (but MTMR14),
a phosphatase domain (mutated in inactives) and a coiled-coil domain (but
MTMR10). In addition, MTMR3 and MTMR4 posses a FYVE domain. Three of these
are associated with genetic diseases; mutation
in MTM1 leads to the Myotubular Myopathy, and mutation in MTMR2 and MTMR13
to the Charcot Marie Tooth disease.
Some interactions between the different members of both subgroups have been
demonstrated; MTM1 interacts with MTMR12 (Nardurkar et al. and Mitchell et
al.) and itself (Schaletzky et al.), MTMR2 with MTMR5 (Dixon et al.), MTMR12
(Nardurkar et al.), MTMR13 (Dixon et al.) and itself (Suter et al.), and MTMR9
with MTMR6 and MTMR7 (Majerous et al.).
